چکیده

Cellulase production study was performed in shake flask and bioreactor system using Cellulomonas cellulans NRRL B 4567 for initial substrate concentration from S0 = 2 to 12 g L . The growth, substrate uptake profile and enzyme activity at different initial substrate concentrations were measured. The results inferred the presence of substrate inhibition kinetics. Various substrate inhibition models were tested and parameters were estimated, using non-linear regression analysis. Han-Levenspiel model was found to be the best fitted model for both shake flask and reactor study. The highest volumetric enzyme activity was observed at initial substrate concentration of S0 = 12 g L –1 and 4 g L in shake flask and bioreactor respectively.

Production of Thermostable α-Amylase and Cellulase from Cellulomonas sp

A bacterium, isolated from rabbit’s waste and identified as Cellulomonas sp., had cellulase and thermostable α-amylase activity when grown on wheat bran. Maximum activity of thermostable α-amylase was obtained by adding 3% soluble starch. However, soybean oil (1 ml l) could increase the production of α-amylase and cellulase in wheat bran. The α-amylase was characterized by making a demonstratio...

Inhibition of Trichoderma Species from Growth and Zoospore Production of Phytophthora Drechsleri and Their Effects on Hydrolytic Enzymes

Understanding the function of Trichoderma species in the control of Phytophthora drechsleri in pistachio orchards is very important.  In this study, the effects of liquid extra-cellular secretions and volatile compounds secreted by 27 isolates of Trichoderma harzianum, T. crassum, T. koningii, T.aureoviride, T. asperellum, T. brevicompactum, T.longibrachiatum and T. virens were investigated on ...

Cellulase Production by Trichoderma reesei using Sugar Beet Pulp

Cellulase production by the fungus Trichoderma reesei was studied using sugar beet pulp (SBP) as a substrate. The subculture medium was a salt solution consisting of KH2PO4, CaCl2, etc. Fungal cells were sub-cultured in an orbital shaker (180 rpm) at 30°C for 1-2 generations (two days for each generation) and were then used as an inoculum. Exponential cells were inoculated into a medium contain...

The prospects of cellulase-producing bacteria for the bioconversion of lignocellulosic biomass

Lignocellulosic biomass is a renewable and abundant resource with great potential for bioconversion to value-added bioproducts. However, the biorefining process remains economically unfeasible due to a lack of biocatalysts that can overcome costly hurdles such as cooling from high temperature, pumping of oxygen/stirring, and, neutralization from acidic or basic pH. The extreme environmental res...

PRODUCTION, RELEASE AND THERMAL CHARACTERIZATION OF CELLULOLYTIC ENZYME FROM CELLULOMONAS sp. STRAIN "0"

Cellulase production by a Cellulomonas sp., isolated in 1985 from forest humus soil along the border of the Caspian Sea in Iran, was investigated. This strain secreted endo-and exo-cellulases in the culture medium, but some of the enzymes produced remained cell membrane bound. Cell bound enzymes were released by various treatments. The highest amount of endo-glucanase (up to 95%) and exo-gl...
